What is sulfadiazine and the related pharmacodynamics?

Sulfadiazine is a sulfonamide antibiotic used in a variety of infections, such as urinary tract infections, trachoma, and chancroid. Sulfonamides are synthetic bacteriostatic antibiotics with a wide spectrum against most gram-positive and many gram-negative organisms. However, many strains of an individual species may be resistant to it.


Sulfonamides inhibit multiplication of bacteria by acting as competitive inhibitors of p-aminobenzoic acid in the folic acid metabolism cycle. Bacterial sensitivity is the same for the various sulfonamides, that is, its resistance to one sulfonamide indicates resistance to all.


Most sulfonamides are readily absorbed orally. Parenteral administration is difficult, since the soluble sulfonamide salts are highly alkaline and irritating to the tissues.


Sulfonamides are widely distributed throughout all tissues. High levels are achieved in pleural, peritoneal, synovial, and ocular fluids. Although these drugs are no longer used to treat meningitis, Cerebrospinal fluid levels are high in meningeal infections. Their antibacterial action is inhibited by pus.

Drug interaction of Sulfadiazine Injection:

Sulfadiazine Injection cannot be used in combination with tetracycline, kanamycin, lincomycin, etc.


① The combination of sulfadiazine and diaminopyrimidine (such as TMP) can produce synergistic effect.


② Some drugs containing p-aminobenzoyl, such as procaine and tetracaine, can produce PABA in vivo. Yeast tablets contain PABA required for bacterial metabolism, which can reduce the effect of this drug, so it is not suitable for combination.


③ It can aggravate nephrotoxicity when used with diuretics such as thiazide or furosemide.

Precautions for Using Sulfadiazine Injection:

① Sulfadiazine Injection can precipitate crystals in the presence of acids, so it should not be diluted with 5% glucose solution.

② Long term or high-dose application of sulfadiazine injection is easy to cause crystalline urine. Sodium bicarbonate should be used at the same time, and a large amount of drinking water should be given to the affected animals.

③ In case of allergic reaction or other serious adverse reactions, stop the drug immediately and give symptomatic treatment.
